ANO9 (TMEM16J) belongs to a family of Ca2+-activated chloride channels and phospholipid scramblases (ANO1 - ANO10 or TMEM16A - TMEM16K). In a genome-wide association study (GWAS), a correlation was found between the expression of the ANO9 variant (ANO9 T604A) and chronic kidney disease (CKD). The function of ANO9 in the kidney and the significance of the ANO9 variant for the development of CKD are currently unclear. However, studies on various kidney cells have shown that ANO9 is a negative regulator of the intracellular calcium signalling pathway. ANO9 is also expressed in human lymphocytes. There, ANO9 modulates the Ca²⁺ influx through ORAI1. Involvement of ANO9 in the development or progression of CKD could reveal new pharmacological targets for the treatment of CKD.
