  • Introduction into the physical principles of protein function in trnacription, RNA interactions, membrane transport and signal transduction,

  • NMR to understand structure and dynamics of proteins

  • High-pressure NMR to investigate the conformational landscape of proteins

  • High-resolution structures by X-ray diffraction and crystallography

  • Time-resolved structures by free electron laser

  • Single particle analysis and cryo-electron microscopy

  • Homology modelling and structure modelling

  • Electron-Tomography, Scanning electron microscopy and correlative light-and electron microscopy 

  • FRET and TIRD microscopy

  • Seminar

  • Presentation and discussion of a research based topic in the field of structural biology with a focus on the methods discussed in the lecture: NMR, X-ray, EM, FRET

Students starting before the winter semester 26/27: Registration is not required for the lectures. There is no exam to sit. However, the content of the lectures may form part of the oral exam.

 

Students starting in the winter semester 26/27 or later: Portfolio (collection of exercises and reflective tasks completed alongside the lectures). Please ask the module coordinator about the requirements. Once it has been finalised, the portfolio must be registered in FlexNow.

The portfolio is not graded.

  • Single Particle Data collection and electron microscopy
  • Two-dimensional and three-dimensional membrane protein crystallization
  • Introduction into EM and X-ray data analysis
  • Homology modelling as tool for structure determination at low resolution
  • Presentation and discussion of own experimental gained data

12Portfolio (graded)*

*The portfolio examination contains following:

  • Internship report of 4.000–10.000 words, including a bibliography, 
  • scientific presentation (15-30 min), 
  • practical scientific work, 
  • data, protocols, and records -including electronic data- from scientific work, prepared in accordance with the principles of good scientific practice. 

The grade of the portfolio is based on the evaluation of the report, the presentation, and the practical work in the lab. The supervisor of the laboratory course can use a template for the evaluation - Evaluation of a practical module - PDF (externer Link, öffnet neues Fenster).
The completed evaluation should then be given to the student as feedback.

Practical Module - Biomolecular NMR-Spectroscopy

  • Collection of 1 and 2 dimensional NMR experiments

  • Analysis of multidimensional NMR spectra

  • Collection and analysis of NMR data to assess protein dynamics

  • Performance of NMR titration data to extract binding characteristics

  • Shift of the conformational equilibrium using high pressure NMR

  • Presentation and discussion of own experimental gained data

Practical Module - Protein Engineering for Superresolution Microscopy

Note! This module is new starting in the 26/27 winter semester and is therefore intended for students who begin their studies in the 26/27 winter semester. However, students who began their studies before the 26/27 winter semester may also take this module. In this case, the modules can be credited toward the Practical Modul “Structure Biology of Membrane Proteins” or the Practical Module “Biomolecular NMR-Spectroscopy”.

Module contents
  • Understanding, development and use of genetically encoded imaging tools (e.g., green fluorescent protein)

  • Mutagenesis as a method for modifying the functional properties of imaging tools

  • Protein purification of imaging tools as a precursor for their in-solution characterization.

  • Spectroscopic in-solution characterization of imaging tools

  • Special focus on photoswitching proteins as tools for e.g. super-resolution microscopy

  • Presentation and discussion of own experimental gained data
